Indications/Uses
Duodenal & gastric ulcer, NSAID-associated gastric acid & duodenal ulcers or erosions, H pylori eradication in PUD, reflux oesophagitis, symptomatic GERD, acid-related dyspepsia & Zollinger-Ellison syndrome.
Dosage/Direction for Use
Adult Acid aspiration prophylaxis 40 mg IV infusion 1 hr before surgery. Severely ill patient w/ either reflux oesophagitis, duodenal or gastric ulcer 40 mg IV infusion once daily for up to 5 days. Zollinger-Ellison syndrome Initially 60 mg IV daily. Doses >60 mg daily to be given as divided doses bd. Hepatic impairment 10-20 mg daily.
Contraindications
Hypersensitivity. Not to be administered w/ atazanavir.
Special Precautions
Exclude gastric malignancy prior to therapy. Lactation. Childn.
Adverse Reactions
GI disturbances eg, diarrhoea, constipation, abdominal pain, nausea, vomiting, flatulence; skin rash, pruritus, urticaria; paresthesia, dizziness, lightheadedness, feeling faint; reversible mental confusion, agitation, depression, hallucinations.
Drug Interactions
Reduced absorption of ketoconazole or itraconazole. Prolonged elimination of diazepam, phenytoin, warfarin. Increased plasma conc of clarithromycin. Increased digoxin bioavailability. Reduced atazanavir exposure. Increased serum levels of tacrolimus.
